簡介:分類號學(xué)號2009004//L丫L/I/IZ/HL/14T//L/14/LL/L/。/LLL/19LLLLH8IHLL6/L//LL/泰山壓虧弦碩士學(xué)位論文論文題目應(yīng)用蛋白質(zhì)組學(xué)方法篩選乳腺癌相關(guān)蛋白質(zhì)作者姓名侯巍巍學(xué)科、專業(yè)病理學(xué)與病理生理學(xué)。學(xué)位類型科研學(xué)位型研究方向腫瘤方向指導(dǎo)教師侯剛教授入學(xué)時間2010年9月論文工作起止時間2011年10月2013年2月應(yīng)用蛋白質(zhì)組學(xué)方法篩選乳腺癌相關(guān)蛋白質(zhì)研究生侯巍巍專業(yè)病理學(xué)與病理生理學(xué)導(dǎo)師侯剛教授中文摘要目的本實(shí)驗(yàn)運(yùn)用雙向凝膠電泳TWODIMENSIONALGELELECTROPHORESIS,2DE技術(shù)、質(zhì)譜分析MASSSPECTROMETRY,MS技術(shù)以及生物學(xué)信息數(shù)據(jù)庫搜索等方法,篩選乳腺癌組織與周圍正常乳腺組織之間的差異蛋白,通過比較這些差異表達(dá)蛋白譜,尋找和鑒定與乳腺癌相關(guān)的蛋白質(zhì),深入了解這些蛋白質(zhì)的結(jié)構(gòu)和功能,篩選出與乳腺癌診斷及預(yù)后相關(guān)的特異性標(biāo)志物并為進(jìn)一步研究奠定基礎(chǔ)。方法選取新鮮乳腺浸潤性導(dǎo)管癌組織及癌旁30CM以外的周圍乳腺正常組織經(jīng)病理證實(shí)30例進(jìn)行雙向凝膠電泳,得到的差異蛋白點(diǎn)用IMAGEMASTER2DPLATINUM50圖像分析軟件進(jìn)行分析,顯示蛋白點(diǎn)強(qiáng)度比值20,說明蛋白點(diǎn)具有表達(dá)差異。運(yùn)用基質(zhì)輔助激光解吸電離時間飛行質(zhì)譜分析MATRIXASSISTEDLASERDESORPTIONIONIZATIONTIMEOFFLIGHTMASSSPECTROMETRY,MALDITOFMS對所篩選的差異蛋白點(diǎn)做質(zhì)譜鑒定;根據(jù)檢索軟件MASCOT計(jì)算得到蛋白的分值,分值達(dá)到70分以上認(rèn)為這個蛋白質(zhì)鑒定可靠;將質(zhì)譜分析得到的數(shù)據(jù)輸入NCBI數(shù)據(jù)庫來獲得蛋白質(zhì)的信息。結(jié)果利用IMAGEMASTER2DPLATINUM50凝膠圖象分析軟件對兩樣本總蛋白進(jìn)行分析,尋找差異蛋白位點(diǎn),得出結(jié)果浸潤性導(dǎo)管癌組織總共有L0404L5個蛋白位點(diǎn),癌旁正常乳腺組織共得到了100317個蛋白位點(diǎn)。根據(jù)IMAGEMASTER2DPLATINUM50凝膠圖象分析系統(tǒng)分析后所得到的差異蛋白數(shù)
