2、 Physics, Study field: Biologic Physics, Entrance Year: 2006 Abstract Raman tweezers is an optical technology coupled optical tweezers and Raman spectroscopy, also called the Raman spectroscopy co
3、mbined with optical trap (RSOT) technique or the laser tweezers Raman spectroscopy (LTRS) system. A laser beam is introduced into an inverted microscope through a high numerical aperture objecti
4、ve to form an optical trap termed optical tweezers. The optical tweezers can trap a single particle in three dimensions nearly in the balance of the focus of laser beam. The same laser beam
5、 is used to excite Raman scattering of the trapped particle. The scattering light from the particle is collected by the objective and coupled into a spectrograph. With the analysis of the Ra
6、man spectra data, different particles or cells can been recognized and distinguished. It is one of the hot tools on single cell-molecular level in biology field. Microfluidic chips are molecul
7、ar analytical tools for solving the complex analytical challenges associated with the field of biology. Last decade, microfluidic chips have been advanced sharply, which can be named micro- total analysis
8、systems (µ- TAS), lab- on- a- chip or integrated chemistry lab and so forth. Under ideal condition, µ- TAS can intergrate very course of conventional chemical experiments in the laboratory
9、 in the same microdevice, which also is a recent technology in current science study. Microfluidic Raman tweezers combined these two techniques offers a new tool for manipulation and identification of singl
10、e biological cells and microscopic particles automatically. This thesis includes six chapters. Chapter 1 is an introduction, including the history, theory, application and current study of Raman tweeze
11、rs. The main contain is the theories of Raman spectroscopy and optical tweezers, and the application of their combination in biology. The second chapter is the study base of microfluidic chips
12、, mainly containing basic character of microsize, the material and facture of microfluidic chips, and the facture, placement and cleanout of the chip of our laboratory. The third chapter is th
13、e device of Raman tweezes, including laser source, detector, coupled optical path, and so on. The fourth chapter is the capability test of Raman tweezers, including the experiments of rat single cells
14、and polystyrene beads. The fifth chapter is the advance and integration of Raman tweezers, mainly introducing the integration and application of microfluidic Raman tweezers. The sixth chapter is
